Glycerine (Glycerol) for Mushroom Culture

Glycerine is a clear, colourless polyol widely used in mycology as a cryoprotectant and spore‑suspension stabiliser. When added to liquid culture or spore stocks, it reduces ice‑crystal formation, improves long‑term viability and keeps solutions homogeneously mixed.

Why Use Glycerine?

• Cryo‑storage: 10 % v/v glycerine protects mycelium and spores during ‑20 °C or ‑80 °C freezer storage.
• Stable spore syringes: 2–4 % v/v prevents clumping and extends syringe shelf‑life.
• LC carbon boost: 1 % v/v serves as an easily metabolised carbon source when dextrose or LME are limited.
• Food‑grade (≥ 99.7 %), odourless and colour‑free—no extra variables in your media.

Typical Applications & Rates

Use	Glycerine %	How to prepare
Cryo stock	10 %	Add 1 mL glycerine to 9 mL cooled LC or spore solution, mix, freeze at −20 °C / −80 °C.
Spore syringe	2–4 %	Add 0.2–0.4 mL per 10 mL sterile water before scraping spores.
Liquid culture	1 %	Add 10 mL to 1 L media alongside sugars; sterilise as usual.

Handling & Storage

• Store sealed in a cool, dry place—hygroscopic, will absorb water from air.
• Viscous: warm slightly (≤ 40 °C) for faster pouring if room temperature is cold.
• Sterilise by autoclaving with media or by 0.22 µm filtration for heat‑sensitive additions.
